Fluorophore Triplet State Duty Fraction Tool

Stationary alternating bright/dark dwell approximation; no separate photophysical rates are inferred.

Loading tool…

Description

Return bright-state duty fraction and expected observation totals.

Fluorescence signals combine excitation, emission, detector response and background. An intensity ratio, effective lifetime or anisotropy correction describes a supplied optical model; measured signal fractions and ideal energy-transfer efficiencies are different quantities and need appropriate calibration.

Return bright-state duty fraction and expected observation totals.

When to use this tool

Use Fluorophore Triplet State Duty Fraction when checking the stated fluorescence, lifetime, transfer or polarization correction.

How to use the tool and how it works

  1. Enter brightDwell, darkDwell, totalObservation. Use the units and conventions below.
  2. Run the tool. Correct any format, range, or cross-field input error before interpreting the output.
  3. Read brightFraction, expectedBrightTime, cycles. Use the output convention below.

Return bright-state duty fraction and expected observation totals. Require nonnegative dwell times with positive sum and totalObservation >= 0; brightFraction = bright/(bright+dark), expectedBrightTime multiplies observation, cycles = observation/(bright+dark).1

Limitations and assumptions

  • Cycles is an expected fractional count under fixed dwell assumptions, not detected switching events or a fitted stochastic model.
  • Inputs must satisfy both the field types and the cross-field conditions above. Invalid input causes an error; a successful false flag or documented placeholder is a different result.
  • Numeric calculations use binary floating point unless exact decimal-integer arithmetic is explicitly stated. No hidden measurement tolerance or decimal rounding is added; nonfinite or out-of-contract results are rejected. Numeric signed-zero results are exported as zero.

Alternative or Complementary methods

Use calibrated optical measurements and a complete suitable photophysical model, including background, cross-talk and detector effects, for molecular or kinetic inference.
