Diffusion Frap Mobile Fraction Term Tool

Simple background-corrected FRAP intensity normalization without ongoing photobleaching or reference-region correction; out-of-range fractions remain visible.

Description

Return a FRAP intensity-based mobile fraction and normalized signals.

Fluorescence signals combine excitation, emission, detector response and background. An intensity ratio, effective lifetime or anisotropy correction describes a supplied optical model; measured signal fractions and ideal energy-transfer efficiencies are different quantities and need appropriate calibration.

Return a FRAP intensity-based mobile fraction and normalized signals.

When to use this tool

Use Diffusion Frap Mobile Fraction Term when checking the stated fluorescence, lifetime, transfer or polarization correction.

How to use the tool and how it works

  1. Enter prebleach, postbleach, plateau, background. Use the units and conventions below.
  2. Run the tool. Correct any format, range, or cross-field input error before interpreting the output.
  3. Read mobileFraction, normalizedPostbleach, normalizedPlateau. Use the output convention below.

Return a FRAP intensity-based mobile fraction and normalized signals. Require prebleach distinct from postbleach and background; mobileFraction = (plateau-postbleach)/(prebleach-postbleach), normalized signals subtract background then divide by prebleach-background.1

Limitations and assumptions

  • Results are not clamped to [0,1]; this does not fit recovery kinetics or correct photofading, transport geometry or sample motion.
  • Inputs must satisfy both the field types and the cross-field conditions above. Invalid input causes an error; a successful false flag or documented placeholder is a different result.
  • Numeric calculations use binary floating point unless exact decimal-integer arithmetic is explicitly stated. No hidden measurement tolerance or decimal rounding is added; nonfinite or out-of-contract results are rejected. Numeric signed-zero results are exported as zero.

Alternative or Complementary methods

Use calibrated optical measurements and a complete suitable photophysical model, including background, cross-talk and detector effects, for molecular or kinetic inference.
