Description
Subtract supplied donor leakage from acceptor signal.
Fluorescence signals combine excitation, emission, detector response and background. An intensity ratio, effective lifetime or anisotropy correction describes a supplied optical model; measured signal fractions and ideal energy-transfer efficiencies are different quantities and need appropriate calibration.
Subtract supplied donor leakage from acceptor signal.
When to use this tool
Use Fluorescence Donor Leakage Term when checking the stated fluorescence, lifetime, transfer or polarization correction.
How to use the tool and how it works
- Enter donorSignal, leakageCoefficient, measuredAcceptor. Use the units and conventions below.
- Run the tool. Correct any format, range, or cross-field input error before interpreting the output.
- Read leakage, correctedAcceptor. Use the output convention below.
Subtract supplied donor leakage from acceptor signal. Require leakageCoefficient >= 0; leakage = donorSignal*coefficient, correctedAcceptor = measuredAcceptor-leakage.1
| Input field | Meaning and units | Accepted input |

} Limitations and assumptions
- The coefficient is not fitted; signed corrected values are retained, without completing FRET or optical calibration.
- Inputs must satisfy both the field types and the cross-field conditions above. Invalid input causes an error; a successful false flag or documented placeholder is a different result.
- Numeric calculations use binary floating point unless exact decimal-integer arithmetic is explicitly stated. No hidden measurement tolerance or decimal rounding is added; nonfinite or out-of-contract results are rejected. Numeric signed-zero results are exported as zero.
Alternative or Complementary methods
Use calibrated optical measurements and a complete suitable photophysical model, including background, cross-talk and detector effects, for molecular or kinetic inference.
