Plating Adjusted Transformation Efficiency
Estimate transformants per microgram from an aliquot plate, accounting for recovery volume and plate dilution.
Description
Transformation efficiency estimates recovered transformants per mass of DNA.
Transformation efficiency estimates recovered transformants per mass of DNA. An aliquot plate must be scaled by recovery volume and its dilution before comparing experiments.1
Estimate transformants per microgram from an aliquot plate, accounting for recovery volume and plate dilution.
When to use Plating Adjusted Transformation Efficiency
Use this tool to estimate transformants per microgram from an aliquot plate, accounting for recovery volume and plate dilution.
Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
How to use Plating Adjusted Transformation Efficiency and understand its results
- Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
- Enter transformantColonies, dnaMicrograms, recoveryVolumeMl, platedVolumeMl, dilutionFraction using the field table and calculation rules, then run the tool.
- Read estimatedTotalTransformants, transformantsPerMicrogram. The explanation below defines the units, signs, ordering and any flags or placeholder values.
- If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
| Input field | Meaning | Value type; see rules below | Requirement |
|---|---|---|---|
| transformantColonies | Transformant colonies | integer; minimum: 0 | Required |
| dnaMicrograms | Dna micrograms | number | Required |
| recoveryVolumeMl | Recovery volume ml | number | Required |
| platedVolumeMl | Plated volume ml | number | Required |
| dilutionFraction | Dilution fraction | number; minimum: 0; maximum: 1 | Required |
Enter nonnegative integer transformant colonies, positive DNA mass in micrograms and positive recovery/plated volumes in mL. Plated volume cannot exceed recovery volume and dilution fraction must be in (0,1]. Return estimated total transformants and transformants per microgram. Zero colonies yields zero estimates.
totalTransformants = colonies × recoveryVolume/(platedVolume × dilutionFraction); efficiency = total/DNAμg Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.
| Output field | Meaning | Value type |
|---|---|---|
| estimatedTotalTransformants | Estimated total transformants | number |
| transformantsPerMicrogram | Transformants per microgram | number |

The estimate assumes a representative homogeneous aliquot and correct dilution. It does not verify selection, distinguish background colonies or correct plating recovery, and it is not a transformation protocol.
Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.
Alternative or Complementary methods
Use Countable Plate Selection: Select colony-count plates within caller-specified countability limits, retaining low and excessive counts separately. For a complementary task, use Conjugation Transfer Density: Compare observed recipient conversion and donor-time-normalized conjugation events; growth and secondary transfer are not corrected.
