Conjugation Transfer Density

Compare observed recipient conversion and donor-time-normalized conjugation events; growth and secondary transfer are not corrected.

Description

Recipient conversion and donor-time-normalized transfer density summarize different denominators in a conjugation observation.

Recipient conversion and donor-time-normalized transfer density summarize different denominators in a conjugation observation. They should not be conflated with a corrected intrinsic transfer-rate constant.1

Compare observed recipient conversion and donor-time-normalized conjugation events; growth and secondary transfer are not corrected.

When to use Conjugation Transfer Density

Use this tool to compare observed recipient conversion and donor-time-normalized conjugation events; growth and secondary transfer are not corrected.

Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.

How to use Conjugation Transfer Density and understand its results

  1. Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
  2. Enter transconjugants, assessedRecipients, donorCells, incubationHours using the field table and calculation rules, then run the tool.
  3. Read recipientConversionFraction, transferEventsPerDonorHour. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement

assessedRecipientsAssessed recipientsinteger; minimum: 1Required
donorCellsDonor cellsinteger; minimum: 1Required
incubationHoursIncubation hoursnumberRequired

Enter nonnegative integer transconjugants, positive integer assessed recipients and donors, and positive incubation hours. Transconjugants cannot exceed assessed recipients. Return a conversion fraction and events per donor hour; zero transconjugants gives both zero.

conversion=transconjugants/recipients; density=transconjugants/(donors × hours)

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

Growth, secondary transfer, recovery and changing donor populations are not corrected. Counts must describe matching experimental populations and the result is an observed normalization rather than a mechanistic rate fit.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use Plating Adjusted Transformation Efficiency: Estimate transformants per microgram from an aliquot plate, accounting for recovery volume and plate dilution. For a complementary task, use Fluctuation Culture Zero Class: Summarize fluctuation-test culture dispersion and the p0 mutation-event estimate from independent culture counts; not mutation frequency per cell.
