Fluctuation Culture Zero Class
Summarize fluctuation-test culture dispersion and the p0 mutation-event estimate from independent culture counts; not mutation frequency per cell.
Description
Fluctuation cultures summarize resistant-count dispersion across independent cultures.
Fluctuation cultures summarize resistant-count dispersion across independent cultures. The zero-culture fraction supports a p₀ mutation-event estimate, which differs from the fraction of resistant cells.1
Summarize fluctuation-test culture dispersion and the p0 mutation-event estimate from independent culture counts; not mutation frequency per cell.
When to use Fluctuation Culture Zero Class
Use this tool to summarize fluctuation-test culture dispersion and the p0 mutation-event estimate from independent culture counts; not mutation frequency per cell.
Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
How to use Fluctuation Culture Zero Class and understand its results
- Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
- Enter resistantCounts using the field table and calculation rules, then run the tool.
- Read meanResistantCount, sampleVariance, varianceMeanRatio, zeroCultureFraction, zeroClassMutationEvents. The explanation below defines the units, signs, ordering and any flags or placeholder values.
- If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
| Input field | Meaning | Value type; see rules below | Requirement |
|---|---|---|---|
| resistantCounts | Resistant counts | list of number | Required |
Enter at least two nonnegative integer culture counts containing both zero and nonzero outcomes. Return mean, sample variance, variance/mean, zero-culture fraction and zero-class mutation events. All-zero and all-positive samples are errors because the finite estimate requires a fraction strictly between zero and one.
sampleVariance=Σ(count−mean)²/(n−1); mutationEvents=−ln(zeroFraction) Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.
| Output field | Meaning | Value type |
|---|---|---|
| meanResistantCount | Mean resistant count | number |
| sampleVariance | Sample variance | number |
| varianceMeanRatio | Variance mean ratio | number |
| zeroCultureFraction | Zero culture fraction | number |
| zeroClassMutationEvents | Zero class mutation events | number |

} Limitations and assumptions
Independent comparable cultures and the zero-class model are assumptions. Cell division totals, selection recovery, mutation rate per cell and full fluctuation-test likelihood are not computed.
Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Lists are limited to 100,000 entries; the rules above may require smaller limits. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.
Alternative or Complementary methods
Use Conjugation Transfer Density: Compare observed recipient conversion and donor-time-normalized conjugation events; growth and secondary transfer are not corrected. For a complementary task, use Stationary Phase Window Audit: Screen the final growth-curve window for a caller-defined relative stability band, rather than equating one count with carrying capacity.
