Delta-Delta-Ct Fold Change 2^-ddCt Calculator
Compute 2^(−ΔΔCt) from an independently normalized ΔΔCt under equal 100-percent amplification efficiency; no reference-gene selection or raw Ct normalization.
Description
Compute 2^(−ΔΔCt) from an independently normalized ΔΔCt under equal 100-percent amplification efficiency; no reference-gene selection or raw Ct normalization.
The common 2-to-the-minus-delta-delta-Ct expression model converts a normalized cycle difference into a conditional fold change. Its amplification-efficiency assumption belongs with the number.
When to use this tool
Compute 2^(−ΔΔCt) from an independently normalized ΔΔCt under equal 100-percent amplification efficiency; no reference-gene selection or raw Ct normalization.
- Prepare the supplied deltaDeltaCt fields for this operation.
- Compare the complete example with the representation used by your own input before carrying the result into another step.
How to use the tool and how it works
- Enter deltaDeltaCt. Use the exact text syntax, units and index conventions explained below.
- Run the tool. If input validation fails, correct the supplied fields before interpreting any result.
- Read result.conditionalFoldChange, result.assumption. Interpret false, null and empty lists using the distinctions below; they can be valid outputs.
Return 2 raised to negative deltaDeltaCt and the explicit equal-100-percent-PCR-efficiency assumption. Zero gives one, positive deltaDeltaCt gives a smaller fold change and negative values give a larger one.1
| Input field | Meaning and representation | Accepted input |
|---|---|---|
| deltaDeltaCt | delta delta ct. | Finite number |
| Output field | Interpretation |
|---|---|
| result | Object containing the named outputs below. |
| result.conditionalFoldChange | The conditional fold change value under the stated formula or text convention. |
| result.assumption | The assumption value under the stated formula or text convention. |

} Limitations and assumptions
- The tool does not calculate deltaDeltaCt from raw samples or validate reference genes and efficiencies. Extreme inputs producing nonfinite output are rejected; measurement uncertainty is not propagated.
- Every declared top-level field is required and unexpected fields are rejected. Numeric inputs must be finite; ordinary text fields are bounded to 8,192 UTF-16 code units, with tighter operation-specific limits where stated.
- Ordinary string lists have at most 128 entries of at most 128 code units each; numeric lists and matrices have separate bounded size checks. Nonfinite results and excessively large serialized outputs are rejected.
- Indexes are zero-based and spans have an exclusive end unless explicitly stated otherwise. A valid false, null or empty-list result is distinct from malformed input.
Alternative or Complementary methods
Use a complete assay-analysis workflow for normalization, quality control and inference. Keep supplied measurements, fixed educational windows and caller-chosen thresholds distinct; these tools perform the documented calculation or retrieve a short reference note.
