Light-Sheet Phototoxicity

Compare light-sheet and conventional illumination dose as a phototoxicity proxy.

Description

Illumination dose compares irradiance-time products for conventional and light-sheet imaging.

Illumination dose compares irradiance-time products for conventional and light-sheet imaging. Dose reduction is a phototoxicity proxy rather than measured damage.1

Compare light-sheet and conventional illumination dose as a phototoxicity proxy.

When to use Light-Sheet Phototoxicity

Use this tool to compare light-sheet and conventional illumination dose as a phototoxicity proxy.

Prepare the optical measurements, geometry, fitted coefficients or caller-selected thresholds requested. Confirm wavelength, radius/diameter, pulse-width, angle and unit conventions before running the calculation.

How to use Light-Sheet Phototoxicity and understand its results

  1. Prepare the optical measurements, geometry, fitted coefficients or caller-selected thresholds requested. Confirm wavelength, radius/diameter, pulse-width, angle and unit conventions before running the calculation.
  2. Enter conventionalIrradianceWPerCm2, conventionalExposureSeconds, lightSheetIrradianceWPerCm2, lightSheetExposureSeconds using the field table and calculation rules, then run the tool.
  3. Read conventionalDoseJPerCm2, lightSheetDoseJPerCm2, doseReductionPercent. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement
conventionalIrradianceWPerCm2Conventional irradiance (W/cm²)number; minimum: 0Required
conventionalExposureSecondsConventional exposure (s)number; minimum: 0Required
lightSheetIrradianceWPerCm2Light-sheet irradiance (W/cm²)number; minimum: 0Required
lightSheetExposureSecondsLight-sheet exposure (s)number; minimum: 0Required

Enter nonnegative irradiances/times with positive conventional dose. Return both J/cm² doses and signed reduction.

dose=irradiance×time; reduction%=100(1−lightSheetDose/conventionalDose)

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

Photochemistry, oxygen, wavelength and cellular damage are not modeled.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use Photobiomodulation Dose: Calculate delivered energy and fluence for photobiomodulation. For a complementary task, use Two-Photon Depth Comparison: Compare measured two-photon and confocal imaging depths.
