Stain Reference Channel Factor Tool

Signed image-channel normalization with explicit reference values, not stain classification or laboratory instructions.

Description

Subtract blank from signal and multiply by nonnegative suppliedReferenceLevel/positive referenceChannel.

Stain analysis distinguishes measured intensity, blank correction, optical density, and linear channel coordinates. These are different representations whose interpretation depends on calibration and the supplied stain model.

Subtract blank from signal and multiply by nonnegative suppliedReferenceLevel/positive referenceChannel.

When to use this tool

Use Stain Reference Channel Factor when you need one explicit stain-signal component.

How to use the tool and how it works

  1. Enter signal, referenceChannel, suppliedReferenceLevel, blank. Use the units and conventions below.
  2. Run the tool. Correct any format, range, or cross-field input error before interpreting the output.
  3. Read normalized, factor. Use the output convention below.

Subtract blank from signal and multiply by nonnegative suppliedReferenceLevel/positive referenceChannel. Return normalized signal and the multiplicative factor.1

Limitations and assumptions

  • Signals share a compatible scale; signed normalized values are retained and the reference is supplied rather than validated or fitted.
  • Inputs must satisfy both the field types and the cross-field conditions above. Invalid input causes an error; a successful false flag or documented placeholder is a different result.
  • Numeric calculations use binary floating point unless exact decimal-integer arithmetic is explicitly stated. No hidden measurement tolerance or decimal rounding is added; nonfinite or out-of-contract results are rejected. Numeric signed-zero results are exported as zero.

Alternative or Complementary methods

Review the complete imaging and stain calibration, deconvolution model, and appropriate professional interpretation separately.
