Biophysics: FRET Backgroundsubtracted Ratio Tool

Compute a background-subtracted apparent FRET acceptor fraction only; excludes spectral bleedthrough, gamma correction, distances and efficiency calibration.

Description

Return blank-corrected donor/acceptor signals and acceptor fraction.

Fluorescence signals combine excitation, emission, detector response and background. An intensity ratio, effective lifetime or anisotropy correction describes a supplied optical model; measured signal fractions and ideal energy-transfer efficiencies are different quantities and need appropriate calibration.

Return blank-corrected donor/acceptor signals and acceptor fraction.

When to use this tool

Use Biophysics: FRET Backgroundsubtracted Ratio when checking the stated fluorescence, lifetime, transfer or polarization correction.

How to use the tool and how it works

  1. Enter donor, acceptor, donorBackground, acceptorBackground. Use the units and conventions below.
  2. Run the tool. Correct any format, range, or cross-field input error before interpreting the output.
  3. Read result. Expand result to inspect the returned fields.

Return blank-corrected donor/acceptor signals and acceptor fraction. Require both corrected signals nonnegative and their sum positive; acceptorFraction = correctedAcceptor/(correctedAcceptor+correctedDonor).1

Limitations and assumptions

  • This simple intensity fraction is not a fully corrected FRET efficiency or distance; leakage and detector response are not modeled.
  • Outputs are nested inside result. Inputs must satisfy both the field types and the cross-field conditions above. Invalid input causes an error; a successful false flag or documented placeholder is a different result.
  • Numeric calculations use binary floating point unless exact decimal-integer arithmetic is explicitly stated. No hidden measurement tolerance or decimal rounding is added; nonfinite or out-of-contract results are rejected. Numeric signed-zero results are exported as zero.

Alternative or Complementary methods

Use calibrated optical measurements and a complete suitable photophysical model, including background, cross-talk and detector effects, for molecular or kinetic inference.
