Bioanalyzer Concentration
Interpolate sample concentration from a caller-supplied Bioanalyzer ladder peak.
Description
A peak-area ratio can scale a sample to a supplied ladder concentration.
A peak-area ratio can scale a sample to a supplied ladder concentration. This assumes proportional signal response for comparable peaks.1
Interpolate sample concentration from a caller-supplied Bioanalyzer ladder peak.
When to use Bioanalyzer Concentration
Use this tool to interpolate sample concentration from a caller-supplied Bioanalyzer ladder peak.
Prepare the upstream counts, expression values, sequences or record lists requested by this tool. Keep corresponding features in the same order and confirm count units, transformations and caller-selected criteria.
How to use Bioanalyzer Concentration and understand its results
- Prepare the upstream counts, expression values, sequences or record lists requested by this tool. Keep corresponding features in the same order and confirm count units, transformations and caller-selected criteria.
- Enter samplePeakArea, ladderPeakArea, ladderConcentrationNgUl using the field table and calculation rules, then run the tool.
- Read concentrationNgUl, areaRatio. The explanation below defines the units, signs, ordering and any flags or placeholder values.
- If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
| Input field | Meaning | Value type; see rules below | Requirement |
|---|---|---|---|
| samplePeakArea | Sample peak area | number | Required |
| ladderPeakArea | Ladder peak area | number | Required |
| ladderConcentrationNgUl | Ladder concentration ng ul | number | Required |
Enter nonnegative sample area, positive ladder area and positive ladder concentration ng/µL. Return ratio and scaled sample concentration. Zero sample area gives zero concentration.
areaRatio=sampleArea/ladderArea; concentration=areaRatio×ladderConcentration Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.
| Output field | Meaning | Value type |
|---|---|---|
| concentrationNgUl | Concentration ng ul | number |
| areaRatio | Area ratio | number |

This single-reference scaling does not fit a calibration curve, identify peaks or apply instrument-specific response and dilution corrections. The title does not imply an instrument reading was imported.
Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.
Alternative or Complementary methods
Use DV200 Fragment Quality: Calculate the share of RNA fragments longer than a user-selected cutoff. For a complementary task, use DPCR Poisson Concentration: Convert digital-PCR positive partition fraction to Poisson-corrected concentration.
