Protein Dilution Backcalculation

Back-calculate original protein concentration after dilution.

Description

Dilution back-calculation multiplies measured diluted concentration by a volume-based dilution factor.

Dilution back-calculation multiplies measured diluted concentration by a volume-based dilution factor. A factor of one leaves the concentration unchanged.1

Back-calculate original protein concentration after dilution.

When to use Protein Dilution Backcalculation

Use this tool to back-calculate original protein concentration after dilution.

Prepare the canonical sequence, peptide assignments, fitted assay parameters or upstream measurements requested. Keep corresponding entries aligned, use the stated mass and signal units, and remove FASTA headers before entering sequence text.

How to use Protein Dilution Backcalculation and understand its results

  1. Prepare the canonical sequence, peptide assignments, fitted assay parameters or upstream measurements requested. Keep corresponding entries aligned, use the stated mass and signal units, and remove FASTA headers before entering sequence text.
  2. Enter dilutedProteinMgMl, dilutionFactor using the field table and calculation rules, then run the tool.
  3. Read originalProteinMgMl. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement
dilutedProteinMgMlDiluted protein mg mlnumberRequired
dilutionFactorDilution factornumberRequired

Enter nonnegative diluted protein concentration in mg/mL and strictly positive factor. Read modeled original concentration in the same units.

originalMgMl=dilutedMgMl×dilutionFactor

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

The factor is not checked to be at least one and can represent a caller-defined scaling. Assay calibration, recovery and preparation error are not modeled.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use BCA Blank Correction: Subtract the matched assay blank from BCA absorbance. For a complementary task, use ELISA Four Parameter Inversion: Invert a caller-fitted four-parameter logistic ELISA standard curve.
