Disorder Residue Fraction

Estimate intrinsic-disorder residue composition using a caller-selected residue set.

Description

A disorder-composition proxy counts members of a supplied residue set.

A disorder-composition proxy counts members of a supplied residue set. The chosen set controls this feature without applying a structural prediction model.1

Estimate intrinsic-disorder residue composition using a caller-selected residue set.

When to use Disorder Residue Fraction

Use this tool to estimate intrinsic-disorder residue composition using a caller-selected residue set.

Prepare the canonical sequence, peptide assignments, fitted assay parameters or upstream measurements requested. Keep corresponding entries aligned, use the stated mass and signal units, and remove FASTA headers before entering sequence text.

How to use Disorder Residue Fraction and understand its results

  1. Prepare the canonical sequence, peptide assignments, fitted assay parameters or upstream measurements requested. Keep corresponding entries aligned, use the stated mass and signal units, and remove FASTA headers before entering sequence text.
  2. Enter sequence, disorderPromotingResidues using the field table and calculation rules, then run the tool.
  3. Read disorderResidueCount, disorderResidueFraction. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement
sequenceSequencestringRequired
disorderPromotingResiduesDisorder promoting residuesstringRequired

Enter a canonical sequence and case-sensitive residue-set string. Return selected residue count and fraction. An empty set gives zero.

The shared sequence helper trims outer whitespace and uppercases the sequence. It requires at least one residue from ACDEFGHIKLMNPQRSTVWY. Internal whitespace, FASTA headers, ambiguous letters and modified-residue notation are rejected. Sequence indexes refer to the normalized sequence; individual tools may preserve original strings in their returned lists as explained above.

fraction=count(residues contained in selected set)/sequenceLength

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

The set is not normalized and contains no embedded definition of disorder-promoting residues. Order, context, structure and validated disorder prediction are not included.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Extremely large text inputs are rejected. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use Low Complexity Windows: Detect compositionally biased low-complexity windows. For a complementary task, use Protein Instability Score: Score protein dipeptide instability with caller-provided empirical weights.
