Calibration Backcalculation

Invert a linear calibration curve to estimate sample concentration.

Description

Calibration backcalculation inverts a supplied straight-line response model.

Calibration backcalculation inverts a supplied straight-line response model. It also reconstructs the response as an arithmetic consistency check.1

Invert a linear calibration curve to estimate sample concentration.

When to use Calibration Backcalculation

Use this tool to invert a linear calibration curve to estimate sample concentration.

Prepare the measured profile, formula counts, upstream model outputs or caller-defined reference criteria requested. Keep paired entries aligned and units, sample basis and preprocessing consistent.

How to use Calibration Backcalculation and understand its results

  1. Prepare the measured profile, formula counts, upstream model outputs or caller-defined reference criteria requested. Keep paired entries aligned and units, sample basis and preprocessing consistent.
  2. Enter sampleSignal, calibrationIntercept, calibrationSlope using the field table and calculation rules, then run the tool.
  3. Read backCalculatedConcentration, reconstructedSignal. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement
sampleSignalSample signalnumberRequired
calibrationInterceptCalibration interceptnumberRequired
calibrationSlopeCalibration slopenumberRequired

Enter finite signal/intercept and nonzero slope, which may be negative. Return concentration and reconstructed response.

concentration=(signal−intercept)/slope; reconstructed=slope×concentration+intercept

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

Concentration may be negative. No calibration fit, uncertainty or valid concentration range is inferred.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use Internal Standard Concentration: Quantify analyte concentration from internal-standard area and caller-provided response factor. For a complementary task, use Detection Limit: Estimate LOD from a measured standard concentration and target signal-to-noise ratio.
