Methylation Standard Interpolation
Linearly interpolate methylation percentage from two assay standards.
Description
Two-standard calibration interpolates a methylation percentage from a supplied signal.
Two-standard calibration interpolates a methylation percentage from a supplied signal.1
Linearly interpolate methylation percentage from two assay standards.
When to use Methylation Standard Interpolation
Use this tool to linearly interpolate methylation percentage from two assay standards.
Prepare the already measured signals, counts, coordinates, fitted coefficients or sequence strings requested. Keep assay definitions, units, aligned lists and caller-selected thresholds consistent.
How to use Methylation Standard Interpolation and understand its results
- Prepare the already measured signals, counts, coordinates, fitted coefficients or sequence strings requested. Keep assay definitions, units, aligned lists and caller-selected thresholds consistent.
- Enter lowerStandardSignal, lowerStandardPercent, upperStandardSignal, upperStandardPercent, sampleSignal using the field table and calculation rules, then run the tool.
- Read interpolationFraction, interpolatedMethylationPercent, withinStandardRange. The explanation below defines the units, signs, ordering and any flags or placeholder values.
- If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
| Input field | Meaning | Value type; see rules below | Requirement |
|---|---|---|---|
| lowerStandardSignal | Lower standard signal | number | Required |
| lowerStandardPercent | Lower standard methylation (%) | number; minimum: 0; maximum: 100 | Required |
| upperStandardSignal | Upper standard signal | number | Required |
| upperStandardPercent | Upper standard methylation (%) | number; minimum: 0; maximum: 100 | Required |
| sampleSignal | Sample signal | number | Required |
Enter distinct finite standard signals, percentages 0–100 and sample signal. Return estimate and within-standards flag for fraction 0–1.
fraction=(sample−signal1)/(signal2−signal1); estimate=percent1+fraction(percent2−percent1) Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.
| Output field | Meaning | Value type |
|---|---|---|
| interpolationFraction | Interpolation fraction | number |
| interpolatedMethylationPercent | Interpolated methylation (%) | number |
| withinStandardRange | Sample lies within standard range | boolean |

Standards need not increase. Extrapolation is not clipped to 0–100; no fit or uncertainty is supplied.
Numeric inputs, where present, must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors. A successfully returned false flag is an evaluated outcome; its meaning is explained above.
Alternative or Complementary methods
Use Single-CpG Methylation: Calculate single-CpG methylation after background correction. For a complementary task, use Global Methylation Proxy: Apply a caller-supplied calibration to a LINE-1 global methylation proxy.
