MeDIP Enrichment
Calculate MeDIP enrichment after volume normalization.
Description
MeDIP fold enrichment compares volume-normalized IP and input signals.
MeDIP fold enrichment compares volume-normalized IP and input signals.1
Calculate MeDIP enrichment after volume normalization.
When to use MeDIP Enrichment
Use this tool to calculate MeDIP enrichment after volume normalization.
Prepare the already measured signals, counts, coordinates, fitted coefficients or sequence strings requested. Keep assay definitions, units, aligned lists and caller-selected thresholds consistent.
How to use MeDIP Enrichment and understand its results
- Prepare the already measured signals, counts, coordinates, fitted coefficients or sequence strings requested. Keep assay definitions, units, aligned lists and caller-selected thresholds consistent.
- Enter immunoprecipitateSignal, inputSignal, ipVolumeFraction, inputVolumeFraction using the field table and calculation rules, then run the tool.
- Read normalizedIpSignal, normalizedInputSignal, enrichmentFold. The explanation below defines the units, signs, ordering and any flags or placeholder values.
- If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
| Input field | Meaning | Value type; see rules below | Requirement |
|---|---|---|---|
| immunoprecipitateSignal | MeDIP immunoprecipitate signal | number; minimum: 0 | Required |
| inputSignal | Input DNA signal | number; minimum: 0 | Required |
| ipVolumeFraction | Immunoprecipitate volume fraction | number; minimum: 0 | Required |
| inputVolumeFraction | Input volume fraction | number; minimum: 0 | Required |
Enter nonnegative IP, positive input and positive volume fractions. Return normalized signals and fold.
fold=(IP/IPVolumeFraction)/(input/inputVolumeFraction) Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.
| Output field | Meaning | Value type |
|---|---|---|
| normalizedIpSignal | Normalized IP signal | number; minimum: 0 |
| normalizedInputSignal | Normalized input signal | number; minimum: 0 |
| enrichmentFold | MeDIP enrichment fold | number; minimum: 0 |

Fractions are not capped at one. No antibody specificity or percent methylation is inferred.
Numeric inputs, where present, must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.
Alternative or Complementary methods
Use ChIP-qPCR Percent Input: Back-calculate ChIP-qPCR percent input from Ct values and dilution. For a complementary task, use Input Background Correction: Subtract input background from epigenomic sample signals.
