Interval Time Above MIC

Compute time above a supplied MIC in a monoexponential post-peak concentration interval; no regimen or clinical target is recommended.

Description

An exponentially declining post-peak concentration crosses a supplied MIC after a calculable time.

An exponentially declining post-peak concentration crosses a supplied MIC after a calculable time. Clipping that crossing to one dosing interval gives a model fraction of time above the threshold.1

Compute time above a supplied MIC in a monoexponential post-peak concentration interval; no regimen or clinical target is recommended.

When to use Interval Time Above MIC

Use this tool to compute time above a supplied MIC in a monoexponential post-peak concentration interval; no regimen or clinical target is recommended.

Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.

How to use Interval Time Above MIC and understand its results

  1. Prepare the sample, count, growth or assay measurements requested by this tool. Confirm units, sampling periods and reporting bases; supply independently established comparison criteria where requested.
  2. Enter peakConcentration, mic, halfLifeHours, dosingIntervalHours using the field table and calculation rules, then run the tool.
  3. Read hoursAboveMic, intervalAboveMicPercent. The explanation below defines the units, signs, ordering and any flags or placeholder values.
  4. If the tool reports an input issue, correct the values and run it again. Apply the stated assumptions when interpreting a successfully returned result.
Input fieldMeaningValue type; see rules belowRequirement
peakConcentrationPeak concentrationnumberRequired
micMicnumberRequired
halfLifeHoursHalf life hoursnumberRequired
dosingIntervalHoursDosing interval hoursnumberRequired

Enter positive peak concentration and MIC in matching units, plus positive half-life and interval in hours. The tool calculates the crossing, clips it to 0–interval and returns hours plus interval percentage. A peak at or below MIC yields zero modeled hours above it.

crossingHours=halfLife × log2(peak/MIC); hoursAbove=clamp(crossing,0,interval)

Units, indexing, boundary conditions and exceptional cases are defined in the calculation rules above.

Limitations and assumptions

This is a single post-peak monoexponential model, without absorption, distribution, repeated-dose accumulation or protein binding. It does not choose a regimen, MIC or clinical target.

Numeric inputs must be finite. Follow the calculation-specific rules for overflow, underflow and exceptional cases. Missing required fields, extra fields, wrong value types and violations of the calculation rules are input errors.

Alternative or Complementary methods

Use Growth Inhibition MIC Endpoint: Derive the observed MIC from a monotone dilution-growth series, preserving below- and above-range censoring. For a complementary task, use Observed Decimal Reduction Time: Estimate a measured D value from positive viable counts and exposure time under log-linear death assumptions.
